Types of Immunofluorescence Tests at NPL

Immunofluorescence Test
Direct Immunofluorescence (DIF)
Used for diagnosing vesiculobullous (blistering) skin disorders and kidney diseases.
How it works
A skin or kidney biopsy is taken and stained with fluorescent antibodies that detect specific immune proteins (IgG, IgA, IgM, C3, and fibrin) deposited in tissues. Under a fluorescence microscope, these immune deposits light up, helping diagnose autoimmune conditions.

  • Skin Disorders: Pemphigus vulgaris, Bullous Pemphigoid, Dermatitis Herpetiformis, Lupus Erythematosus etc.
  • Kidney Diseases: Kidney Diseases: Lupus Nephritis, IgA Nephropathy, Anti-GBM Disease etc.
Indirect Immunofluorescence Assay (IFA)
IFA is a gold-standard laboratory technique that detects specific autoantibodies in a patient’s blood sample. It is widely used for diagnosing autoimmune disorders and connective tissue diseases.

How it works
The patient’s serum (blood) is mixed with specially prepared antigen-coated slides. If antibodies against the antigen are present in the blood, they bind to the slide and are detected using fluorescent markers. The fluorescence intensity indicates the presence of specific autoantibodies.
